Development of chimeric gene promoters responsive to hypoxia and ionizing radiation

  • 摘要: 介绍两种应用射线和乏氧应答启动子调节的GDEPT(定向基因酶前体药物治疗)系统。用治疗性、条件性或肿瘤特异性启动子控制GDEPT是一种控制靶基因在肿瘤内表达的方法。通过选择性启动子,使基因靶向肿瘤内表达,提高特异性和靶向性,解决了肿瘤基因治疗中的主要限制。

     

    Abstract: We describe two systems that make use of gene, directed enzyme prodrug therapy, regulated by radiation or hypoxic, responsive promoters. The use of treatment-, condition- or tumor-specific promoters to control gene-directed enzyme prodrug therapy is one such method for targeting gene expression to the tumor. The development of such strategies that achieve tumor targeted expression of genes via selective promoters will enable improved specificity and targeting thereby addressing one of the major limitations of cancer gene therapy.
