Abstract:
Objective To investigate the effect of the knockdown of ilent information regulation factor 2 related enzyme on inflammatory factor IL-6 produced in irradiated mesenchymal stem cells(MSCs) and explore the radioprotective effect of SIRT1.
Methods MSCs was treated with different dose of irradiation, and the expression of IL-6 in different time was detected by Western-blot and RT-PCR; MSCs were divided into control group, radiation group and combination group, the expression of IL-6 and SIRT1 was detected by Western-blot and RT-PCR.
Results Ionizing radiation could lead to the increasing of IL-6 expression in MSCs. In detail, the expression level of IL-6 protein reached the peak in 12 hours after radiation and then slowly recovered to the baseline in 24 hours after radiation. And interfered with SIRT1 could aggravate the expression of ionizing radiation-induced IL-6 and worsen inflammation.
Conclusion SIRT1 may inhibit the ionizing radiation-induced expression of inflammatory cytokines IL-6.
