放射性食管损伤中相关细胞因子及信号通路的研究进展

Research progress on cytokines and signaling pathways related to radiation-induced esophageal injury

  • 摘要: 放射性食管损伤(RIEI)是胸部肿瘤放疗中常见的剂量限制性并发症,其发病机制涉及辐射直接引起的DNA损伤、氧化应激及慢性炎症级联反应等。笔者系统阐述了RIEI的病理进程,重点聚焦关键调控细胞因子肿瘤坏死因子α、白细胞介素(IL)-1、IL-6、IL-8、转化生长因子β(TGF-β)等和信号通路(核因子κB、TGF-β1、TGF-β/Smad、丝裂原活化蛋白激酶等)在RIEI中的多层次互作模式,为开发RIEI的靶向干预策略提供理论依据,也为未来开展精准放射防护研究指明了方向。

     

    Abstract: Radiation-induced esophageal injury (RIEI) is a common dose limiting complication in chest tumor radiotherapy, and its pathogenesis involves direct radiation-induced DNA damage, oxidative stress, and chronic inflammatory cascade reactions. The authors systematically elaborates the pathological progression of RIEI, with a focus on the multi-level interaction mechanisms involving key regulatory cytokines (such as tumor necrosis factor-α, interleukin (IL)-1, IL-6, IL-8, and transforming growth factor β (TGF-β)) and signaling pathways (including nuclear factor-κB, TGF-β1, TGF-β/Smad, and mitogen-activated protein kinase) in RIEI. The authors aims to provid a theoretical basis for developing targeted intervention strategies and also point the way for future research on precision radioprotection.
