Gene therapy of thyroid carcinoma

Abstract:
Normally, differentiated thyroid carcinoma(DTC) is a disease of good prognosis, but about 30% of the tumors are dedifferentiate, which are inaccessible to standard therapeutic procedures such as "operation 131I therapy mid thyroid hormone". Both internal and abroad experts are researching a new therapy of dedifferentiated thyroid carcinoma——gene therapy. Many of them utilize methods of it, but follow different strategies:(1) transduction of the thyroid sodium/iodide transporter gene to make tissues that do not accumulate iodide treatable by 131I therapy;(2) strengthening of the anti-tumor immune response;(3) suicide gene therapy;(4) depression the generation of tumor cells;(5) gene therapy of anti-vascularization.

Abstract:
Recently, molecular imaging technologies detecting cell apoptosis in vivo are developing rapidly. The one of the most promising techniques to detect apoptosis in vivo is radionuclide imaging with radioLabeled annexin V. Due to the non-invasiveness, quantitation and other advantages, radionuclide tracing apoptotic imaging in vivo, which potentially has numerous valuable applications, has been greatly developed. The purpose of this paper is to review the application and development of cell apoptosis imaging with radionuclide tracing techinique.

Abstract:
Brown adipose tissue(BAT) is a kind of normal tissue in healthy subject, which has unique physiological function. BAT will accumulate higher 18F-fluorodeoxyglucoae(18F-FDG) in the process of its being excited, and presents variety of radioactive accumulation while performing PET scan. It will benefit improving diagnostic specificity of 18F-FDG PET imaging to master the regular patterns, features of uptaking 18F-FDG and differential methods.
Investigations of labeling insulin-like growth factor 1 analogue with 99mTc

Abstract:
Objective To establish a useful and stable method for labeling insulin-like growth factor I analogue(IGF-1A) with 99mTc. Methods The "pretinning" method was adopted to label IGF-1A. Several labeling conditions were tested. The volume of Tween 80 was from 0 to 10μl, the labeling efficiency was determined from 0.25 h to 6 h after labeling, SnCl2·2H20 from 0.75g/L to 4g/L, the amount of IGF-1A from 20 to 100μg, ihe volume of 99mTc perrhenate was from 10 to 200μL The in vitro stability of 99mTc-IGF-IA was analyzed by using Iranian serum or sodium chloride as challenging agent, and the labeling efficiency was determined from 1 h to 24 h after added challenging agent. Results The labeling efficiency of 99mTc-IGF-1A could reach(94.43±0.75)% and the mass fraction of radiocolloid was(3.47±0.71)%. It was(85.57±2.81)% after incubation 6h at room temperature, and was(54.07±3.86)% after incubation 24 h with human serum. Conclusions The optimum labeling method was 100μl stannous chloride(3g/L) dissolved in sodium gluconate, 40μl ICF-1A(2g/L), 300μl Na3PO4, 2μl 0.1% Tween 80, 50μl 99mTc perrhenate, incubation 0.5h at room temperature, then added 500μl NaH2PO4. This method of labeling IGF-1A with 99mTc using SnCl2·2H2O and sodium gluconate was stable and high labeling efficiency was obtained.

Abstract:
Objective To investigate a fine experiment condition of indirect labeling method that Hepama-1-SPIONs(supeiparamagnetic iron oxide nanoparticles)was labelled with rhenium-188 complex([188Re(CO)3(H2O)3]+). Methods The Hepama-1-SPIONs bridged by bifunctional chelating agent 2-picolylamine-N,N-diacetic acid(PADA) was labelled rhenium-188-tricarbonyl complex prepared with high labeling yields. Results The labeling rate of [188Re(CO)3(H2O)3]+-PADA-Hepama-1-SPIONs was more than 80%, and was extremely stable in vitro, The labeling rate of 188Re-(CO)3(H2O)3-PADA-Hepama-1-SPIONs was more than 90% after 48 in calf serum and isotonic Na chloride. Conclusions 188Re-(CO)3(H2O)3-PADA-Hepama-1-SPIONs shows it's fine stability and biological properties, and it is well worth further researching as bio-magnetically targeted radiotherapy for cancer.
Experimental study of capacity of the blood blow of spinal cord using radiolabelling microspbere technique

Abstract:
Objective In order to study the relation ship between local blood flow and neurofunction at the Later stage of spinal injury. Method Measured the capacity of the local blood flow in the spinal cord injury by the radiolabelling microsphere technique and scored the nervus function grade. Result There are no correlation between the local blood flow and the nervus function. Conclusion It is obvious that the increase of the local blood supply can't reflect the nervus function.
The dilemma of the target delineation with PET-CT in the radiotherapy planning for malignant tumors

Abstract:
Questions are currently still not clearly conclusively answered, when contouring target with PET/CT in the radiotherapy planning. Who needs to contour the PET volume, the radiation oncologist or the nuclear medicine physician? Which factors will contribute to the increase of the inter-observer variability between them? What should be taken as the optimal SUV threshold to demarcate malignant tumors from the benign tissue? When the PET volume is not in accordance with the CT finding in the local area, which part should be contoured as the target? When PET positive while CT negative or PET negative while CT positive were shown in the regional lymph node draining area or in the remote region, in which way the target could be decided and selected? Further study concerning the corresponding relationship between PET-CT and its registered pathological specimen are needed. The long-term clinical results of increased therapeutic ratio will finally verify the applicability of the guideline to contour the target with PET-CT in the radiotherapy planning.
The value of retinoic acid-induced redifferentiation in the treatment of differentiated thyroid carcinoma

Abstract:
About 30% of differentiated thyroid carcinoma(DTC) loses its ability of differentiation, which results in poor or no effect of the radioiodine therapy and the thyroid hormone replacement therapy after operation. Retinoic acids, the biologically active metabolite of vitamin A, is of central importance for growth, proliferation and differentiation of various cells. In this review, summarized the mechanism of the dedifferentiation among the DTC, the retinoic acids-induced redifferention and the clinical use of retinoic acids. It is demonstrated by many studies that retinoic acids-induced redifferentiaon has a large potential in the treatment of DTC.
The observation of curative effects by therapy with low-dose 131I in younger with Graves' disease

Abstract:
Objective To observe the the curative effects in younger with Graves disease therapied by 131I. Methods The dose of 131I is administrated with 1480~2220kBq/g of thyroid tissue which was decided by many factors that include the patient's Age, volume of thyroid, course and if antihyroid drug is administrated. The curative effects was classfide into four groups: complete remission, excellence, parts of remission, no effect. Results 47 were complete remission, 34 were excellence, 10 were the parts of remission and 0 was no effects. The total effective power was 100%. Conclusions Therapy with low-dose of 131I for younger with Graves' disease is an effect, simple and safe method. Repeating treatment with, 131I will improve the curative rate of Graves' disease in younger, and the incidence of hypothyroidism cannot be increased.

Abstract:
Objectives To investigate the relationship between nasopharyngeal carcinoma hypoxia degree evaluated by imaging of the marker(99m)technetium-labeled 4, 9-diaza-3, 3, 10, 10-tetramethyldodecan-2, 11-dione dioxime(99mTc-HL91) and tumor regression in medium-term radiotherapy. Methods 34 patients with nasopharyngeal carcinoma staging from Ⅱ-IVa underwent 99mTc-HL91 SPECT imaging, integrated CT scan and its image fusion one week before receiving radical and continuous radiotherapy. Tumor/muscle, tumor/cerebra of the nasopharyngeal carcinoma and lymph node/muscle, lymph node/cerebra of the neck lymph node metastases were calculated, and lesion regression degree was measured in medium-term radiotherapy. Results ① The diameter of nasopharyngeal carcinoma with positive hypoxia imaging was(1.70±0.68)cm. There was a positive relationship with tumor/muscle and tumor/cerebra(r was 0.607 and 0.640, P was 0.001 and 0.000). ②There was significant difference between tumor/muscle and tumor/cerebra(P was 0.004 and 0.001) of nasopharyngeal carcinoma with grade I and grade ID regression. The regression degree of nasopharyngeal carcinoma in medium-term radiotherapy was negative correlated to the hypoxic degree(tumor/muscle:r=-0.602; tumor/cerebra:r=-0.643, P<0.01), ③99mTc-HL91 imaging detected 28 neck metastasis lymph, nodes. There was no significant line trend between lymph node/muscle, lymph node/cerebra of Lymph node metastases and its size, lesion regression degree in medium-term radiotherapy. Conclusions The Larger the nasopharyngeal carcinoma, the higher the hypoxia degree of lesion, and the less regression in medium-term radiotherapy. There was no significant relationship between hypoxia of neck lymph node metastases and its size and medium-term regression.

Objective To evaluate the value of(18F-fluorodeoxyglucose, 18F-FDG)coincidence imaging and(99mTc-sestamibi, 99mTc-MIBI)myocardial perfusion imaging diagnosis the myocardial viability after myocardial infarct. Methods 24 cases myocardial infarcts patients undertaking angiography, 99mTc-MIBI stress and rest myocardial perfusion imaging, 18F-FDG coincidence metabolism imaging(fasting-glucose stress one day imaging) with single phone emission computer tomography. Results 27 out of 49 99mTc-MIBI reduced myocardial perfusion myocardial segments mismatch fasting and glucose stress 18F-FDG uptake suggesting the viable myocardial. 22 segments with reduced myocardial perfusion match fasting and glucose stress 18F-FDG uptake suggesting no viable myocardial. Fasting imaging viable myocardial can uptake less 18F-FDG but image well, glucose stress imaging viable myocardial can uptake more 18F-FDG and improved the image quality. Conclusions Fasting and glucose stress can show myocardial viability, imaging quality was less well in fasting and improved in glucose stress.

Abstract:
Objective Microarrays was used to text changes of gene expression of thymus cell in C57 mice one moth after exposed to 1 Gy γ-ray irradiation. Methods The difference of gene express between irradiated and un-irradiated mice being analysis by Agilent mouse oligo microarrays. Results In the observed 21319 known genes, 107 up-regulated at least 2-fold(13 excess 4-fold); whereas 9 genes were down-regulated 2-fold(2 more 4-fold). The function of theses genes are known as some basic metabolic process. Conclusions The gene changed in the experiment involved embryo development, organ formation and maintain, immunity or stress, protein compose, apoptosis, signal transduction. The number of gene is much more than that of down-regulated. It ought to be noticed that the gens which coding homy protein in embryo development, and aught also to be noticed that the gens which involve the function of PI3-K.

Abstract:
Objective To explore the molecular mechanism of bone marrow injury of acute irradiation syndrome, gene expression of bone marrow in mice after whole body irradiation was studied. Methods The molecular biology technology including suppression subtractive hybridization, cDNA array hybridization and Northern hybridization were used to study differential gene expression of mRNA level in bone marrow of C57BL/6J mouse 4h after 7Gy 60Co γ-ray radiation in vivo. Results A series possibly induced genes after irradiation were obtained. The function of these genes related to cell cycle control, anti-oxidation, DNA repair, and hematopoiesis-immune response. The differential expression of CDKN1A and S100A8 gene were validated by further experiment. Conclusions The function of those differentially expressed genes after irradiation illuminated that radiation effects, such as DNA damage, cell cycle arrest and oxidation reaction which often happened in mammalian cells, also took place in bone marrow cells after irradiation. Furthermore, the hematopoiesis-immune function and the neuroimmunoendocrine regulation in bone marrow were also changed.

Abstract:
Ataxia-telangiectasia(AT) is caused by the mutation of AT(ATM)mutated gene and it is characterized by high radiosensitivity of AT calls. Progress in the research of AT is reviewed in this paper, including the clinical symptoms of this disorder, as well as the localization, structure and function of ATM gene, and summarized the studies on the gene mutation and high radiosensitivity of AT cells in recent years. Further study to ATM gene may provide an important entrance for radiosensitivity-related therapy.
Comparative study of the influence of radioactive seeds implantation and radiotherapy on cytokine of VX2 tumor rabbits

Abstract:
Objective To discuss the influence of radioactive seeds implantation and radiotherapy on VX2 tumor cytokine. Methods 60 male New Zealand White rabbits were randomly divided into 3 groups, 20 for radioactive seeds implantation group, 20 for radiotherapy group, and 20 for control group. After 12d the implantation with the VX soft tumor, radioactive seeds implantation and radiotherapy were performed separalely, the control group underwent no therapy. IL-2, IL-6, IL-8, TNF and IGF were measured. Results The survival periods of experimental group were 15d longer than the control group. The concentration of IL-2, IL-8 in serum decreased gradually, the decreasing extent control > radiotherapy > radioactive seeds implantation;the concentration of IL-6, IGF raised gradually, rising extent control > radiotherapy > radioactive seeds implantation. Conclusions The immune system of rabbits with VX2 tumor were suppressed, there is little influence of radioactive seeds implantation and radiotherapy on cytokine. Radioactive seeds implantation and radiotherapy are effective methods to treat VX2 tumor.
