Study on 125I-vascular endothelial growth factor receptor-3 polyclonal antibody biod-istribution in rat

Abstract:
Objective To study the biodistribution of 125I-Flt4 PcAb in rat and to establish the foundation of sentinel lymph node (SLN) exceptional orientation. Methods Flt4 PcAb was labeled with 125I via chloramine T method. 125I-Flt4 PcAb was subcutaneous injected in dorsum of foot in rats. The rats were killed at different stages after injection and the popliteal lymph nodes and major organs were taken out, then the radioactivity count of those organs and lymph nodes were measured. Results The labeling efficiency was 46.5%. The specific activity and the radiochemical purity of 125I-Flt4 PcAb were 1.72×1012Bq/g and 96.7% respectively. It showed higher distribution in popliteal lymph nodes and blood than the other organs. The radioactivity ratio of popliteal lymph node to blood was more than 2.5 in 16 hours after injection. Conclusions:(1)It was successful to label Flt4 PcAb with 125I via chloramine T method.(2)Flt4 PcAb would concentrate in lymph node after subcutaneous injection. (3)This study provided experimental basis for SLN exceptional orientation with Flt4 PcAb.
Direct labeling of anti β-human chorionic gonadotropin monoclonal antibody with 99mTc

Abstract:
Objective To label the anti-β-human chorionic gonadotropin monoclonal antibody with β. Methods It was direct labeled using SnCl2 as a reduction agent. Results The labeling procedure was finished within 1.5 hours. The labeling efficiency of the labeled anti-β-human chofionic gonadotropin monoclonal antibody was 88.1%-97.5%, 99mTc colloidal was 0.96%-1.27%, 99mTcO4- was 1.1%-9.92%. And it retained its specific biological activation. Conclusion This direct method is simple, rapid and stable, and it hope to be used as radioimmunodiagnosis for gestational trophoblastic turnout.

Abstract:
Objective To investigate a supersensitive for nucleic acid hybridization assay. Methods Using the two-roung enzyme-amplified action, PCR amplification, Tb chelate and time-resolved detection technology, target PSA DNA was detected. Results The range of the standard curve for detecting target PSA DNA is more than two order of magnitude; the sensitivity was 10pmol/L (0.5fmol/well); the accuracy and the precision was 77%~95% and 12.9%~15.3% respectively when the target PSA DNA was 10,30 and 60pmol/L. Conclusion The method is a supersensitive and new assay technology which has a good prospect of applications.

Abstract:
Myopia is the most familiar ametropia. Animal experimental models include form deprivation myopia and defocus myopia. Experimental animals we often use are chicken and mammals. The retinal dopamine system and vision expierence have close relations with the regulation of eyeball's growth after birth, while the change ofdopamine transporter may reflect the change ofdopamine in the synaptic cleft more directly.

Comparison of adenosine stress and exercise stress 201Tl myocardial perfusion imaging for diagnosis of coronary heart disease

Abstract:
Objective The aim of this study was to compare the diagnostic values of adenosine and exercise stress 201Tl myocardial perfusion imaging for detecting coronary heart disease (CHD). Methods 41 patients with suspected CHD were randomly divided into two groups. In one group adenosine stress was submitted, the exercise stress myocardial SPECT was performed in another. Coronary angiography (CAG) was performed in each patient within 2 weeks before or after SPECT. The result of CAG was taken as "gold standard" of CHD. They compared the diagnostic value of two methods. Results In adenosine group, the sensitivity, specificity, positive predictive value, negative predictive value, accuracy are 92.86%, 57.14%, 81.25%, 80.00%, 80.95% respectively. In exercise stress group, are 100%, 60.0%, 71.43%, 100%, 80.00% respectively. Detection rates of coronary artery lesions were 66.67% and 72.22% in two groups respectively. Conclusion Adenosine stress testing and exercise stress testing 201Tl myocardial perfusion imaging may provide similar value for detection of CHD.

Abstract:
Objective To explore the clinical value of PET-CT for the diagnosis of unstable plaques in atherosclerotic lesions. Methods 60 cases underwent whole-body 18F-FDG PET-CT were retrospectively evaluated, including 6 cases with normal control group and 54 cases with atherosclerotic plaques. 54 cases with SUV and CT value in atherosclerotic lesions of large arterial wall were detected retrospectively. 54 high 18F-FDG uptake cases in atherosclerotic plaques were divided into three groups according the CT value:soft plaques group, intermediate plaques group, calcified plaques group. Group 4 was normal. Results In soft plaques group there were 42 sites whose average SUV was 1.553±0.486, in intermediate plaques group there were 30 sites whose average SUV was 1.393±0.296, in calcified plaques group there were 36 sites whose average SUV was 1.354±0.189, in normal control there were 33 sites whose average SUV was 1.102±0.141. The SUV showed significant difference among the four groups (F=678.909, P=0.000). There were significant difference between normal 18F-FDG uptake group and high 18F-FDG uptake groups. Conclusions 18F-FDG had different degree high uptake in large atherosclerotic plaques, the soft plaques had the highest 18F-FDG uptake among them. The results suggested that 18F-FDG PET-CT had significant potential valuable in the diagnosis of unstable soft plaques in atherosclerotic lesions.
Value of captopril intervened 99mTc-ethylenedicysteine renal dynamic imaging in renovascular hypertension patients

Abstract:
Objective To show the value of captopril renal scintigraphy for detecting a renovascular cause in hypertensive patients and to assess the ability to predict the beneficial effect of revascularization on renal function. Methods Fifty-five consecutive patients with suspected renovascular hypertension (RVH) underwent renal scintigraphy after injection of 99mTc ethylenedicysteine (99mTc-EC) after oral administration of 50 mg captopril given 60 min before the test. Baseline scintigraphy was performed 24 h later if the 99mTc-EC results were abnormal. Results 20 patients had renal artery stenosis(RAS) in 22 patients whom scintigraphy had a high probability of RVH. The result was indeterminate in 10 patients, 5 of them had RAS. In 23 patients found a low probability RVH. In 18 of 22 patients with a high probability RVH by scintigraphy, revascularization procedures were performed and 16 patients had beneficial effects, 1 of 4 patients with intermediate RVH had beneficial effects. Conclusion Captopril renal scintigraphy can detect and can predict the beneficial effect of revascularization in some cases.
The value of regional wall motion abnormalities on gated mycardiac perfusion imaging in predicting angiographic stenoses of coronary artery

Abstract:
Objective To determine the possible level of angiographic stenoses of coronary artery at which reversible regional wall motion abnormalities (RWMA) are present on 99mTc-sestamibi (99mTc-MIBI)-gated myocardial perfusion imaging (MPI). Methods ninty patients undergoing coronary angiography MPI within two weeks were recruited. A five grades and nine segments marking system was introduced to assess the RWMA and thickening of left ventricles. Results The sensitivity of reversible RWMA for detecting ≥75% angiographic stenoses was 64%,with a specificity of 95% and positive predictive value of 97%.The presence of reversible RWMA was able to stratify patients with severe angiographic stenoses of 75% or more from those less than 75% with high positive predictive value. A good correlation was noted between the presence of reversible RWMA and the coronary artery jeopardy score. Multivariate analysis showed that the post-stress RWMA and reversible RWMA scores and positive dipyridamole-stress exercise electrocardiogram(ECG) were significant predictors of angiographic severity. Conclusions Reversible RWMA, as shown by dipyridamole stress 99mTc-MIBI MPI, is a significant predictor of angiographic disease with very high specificity and adds incremental value to MPI for the assessment of angiographic severity.
Clinical analyses of radionuclide bone imaging with 132 gastric cancer

Abstract:
Objective To investigate the characteristics and regularity of skeletal metastases with gastric cancer of different pathological types and clinical stage. Methods Radionuclide bone imaging was analysed retrospectively in 132 cases of gastric cancer. And discussing in different pathological types, clinical stage, the number of bone metastasis and distribute sites. Results Total skeletal metastatic rate of gastric cancer was 15.9%(21/132). Multiple bone metastasis 90.5%(19/21). The ratio of bone metastasis of Ⅲ, Ⅳ stage is 18.4%, 26.6% respectively. Low deferential and undifferentiated adenocarcinoma is 15.2%, 20% respectively. Spine bones were the frequently involved metastatic sites, Thoracic and pelvis were the next. Conclusions Keletal metastases with gastric cancer is not frequet. Metastatic sites is widespread and late stage. Radionuclide bone imaging of regularly is useful for diaguosising, treating gastric cancer.
A comparision between adrenomedullin, endothelin and angiotensin Ⅱ in patients with congestive heart failure

Abstract:
Objective To observe the changes of plasma adrenomedu (AM), endothelin (ET) and angioteusin Ⅱ (Ang Ⅱ) in patients with congestive heart failure(CHF). Methods Plasma AM, ET and Ang Ⅱ levels were determined by radioimmuncassay in 95 patients with CHF and 50 healthy control subjects. In 95 patients with CHF, the left ventricular ejection fraction (LVEF) of 56 patients was measured by echocardiography. Results The plasma AM levels were significantly higher in patients with CHF than that in control subjects. The increase of plasma AM, ET and Ang Ⅱ was parallel with the severity of CHF. There was a significant correlation between plasma levels of AM and ET (r=0.7218, P<0.01) and Ang Ⅱ (r=-0.6566, P<0.001). LVEF inversely correlated(r=-0.6258, P<0.001) with plasma AM levels and significantly positive correlated between the plains AM levels and the cardiothoracic ration(r=-0.6037, P<0.05) in patients with CHF. Conclusions These results suggest that AM might play a certain role in the pathophysiology of CHF. The elevated plasma ET and Ang Ⅱ may be responsible for the rise of plasma AM levels in patients with CHF.
Changes of serum and urine β2-microglobulin of leukopenia in hyperthyroidism of Graves' disease before and after 131Ⅰ therapy

Abstract:
Objective To investigate the value of 131Ⅰ therapy and the changes of the serum, urine contents of β2-microglobulin(β2-MG)in leukopenia in patients with Graves' disease before and after 131Ⅰ therapy. Methods Serum and urine β2-MG contents (with radioimmunoassay), serum free triiodothyronine (FT3) and free tetraiodothyronine (FT4) lever (with chemiluminescence immunoassay) were determined before and after 131Ⅰ therapy. At the same time, measure blood ured nitrogen (BUN) and creatine (Cr) Results The cure rate, improvement, invalid, hypothyroidiam and total effective rate of 131Ⅰ therpy are 76.7%, 13.3%, 3.3%, 6.7%, 96.7% respectively. The serum and urine β2-MG contents as well as the serum FT3, FT4 levels in the 30 patients before treatment were significantly higher than those in the controls (P<0.01). While leucocyte was significantly lower than those in the control (P<0.05). After 3 months treatment, their FT3, FT4 levels lower than before treatment (P<0.01),[32-MG levels lower (vs before treatment, P<0.05), leucocyte increased (vs before treatments, P<0.05). After 6 months treatment, their β2-MG,FT3,FT4 levels dropped to approaching normal (vs controls, P>0.05 and vs before treatments, P<0.01). Both BUN and Cr of the controls and before treatment groups have no big difference. The serum FT3, FT4 level was positively correlative to the β2-MG (P<0.05). Conclusions 131Ⅰ therapy is an ideal treatment for Graves' hyperthyroidism, which have many advantages. β2-MG could be used as a diagnostic indicator for leukopenia in patients with Graves' disease.

Abstract:
Objective Observe the relation between endothelin and coronary heart disease around percufaneous coronary intervention (PCI) operation. Methods In hospital the coronary heart disease patients collected blood from the vein in 3 days before PCI operation, the same day, postoperative half an hour or 1 days or 3 days, observe the change of endothelin in plasma. Results There are obvious differences between the endothelin which in the normal control group and 3 days before operative or the same day, having nothing difference with the half an hour postoperative group, the endothelin which in 3 days or 1 day postoperative is obviously higher than the normal control group, the endothelin of half an hour postoperative is obviously lower than foreoporation, having statistics meaning, but 1 or 3 days postoporative is no difference with foreoporative. Conclusions The endothelin is closely with coronary heart disease, but if it could be as judging the index of the PCI postoperative taking a favorable turn and prognosis also needs the further discussion.

Abstract:
The spatial resolution of PET can reach 4-5mm in clinic, but it can not compare with X-ray, CT, MRI and other modality, and consequently restrict the application of PET in some clinical area, especially the inaccurate delineation of target volume in radiotherapy plan. Lots of factors can affect the resolution of PET, one of them is the smear caused by the motion of respiratory. The spatial resolution can be improved after the compensation of respiratory movement in order to enlarge the clinical application of PET. One of the compensation methods is respiratory gating.

Abstract:
Radiotherapy is one of important treatments for glioma. Functional imaging, such as PET, SPECT and MRI, may provide more valuable information not only in display of the evasion extent of glioma but also in demonstration of some biological characteristics of the tumor, such as perfusion, metabolism, hypoxia or proliferation. Thus it may play a role in making an individualized and more exact radiotherapy planning.

Cervical cancer is the common malignancies in woman, 18F-fluorodeoxyglucose (18F-FDG) PET is a well-established method for detecting, staging, cancer recurrence, therapeutic response and prognosis of cervical cancer. PET-CT can accurately locate the anatomical sites of tracer uptake and improve the diagnostic accuraccy of PET.
The protective effect of prostaglandin E1 injection on acute radiation pneumonitis

Abstract:
Objective To explore the protection of prostaglandin E1 injection with radiation-induced lung injury. Methods Thirty-nine female Sprague-Dawley rats were randomly divided into three groups:blank control group, radiation alone group and radiation plus drug group. Irridiation was given to the whole right lung by a single dose of 20Gy with 8MeV electon. Prostaglandin E1 injection 10μg·kg-1·d-1 was injected into the rats of radiation plus drug group by vena candalis per day for five days, starting on the first day. Animal of each group were sacrificed on days 5, 15, 30 respectively after irradiation. Transforming growth factor-β1 (TGF-β1) level in serum was measured and sections of right lung tissues were observed by hematoxylin and eosin staining. Results Pulmonary edema and pneumonia aggravated gradually. TGF-β1 increased significantly beginning from day 5, especially on days 15, 30 (P<0.05) in group radiation and radiation plus drug. TGF-β1 decreased on days 15, 30 (P<0.05) in group radiation plus drug. Conclusions Prostaglandin E1 injection could decrease serum TGF-β1 content. Inhabiting expression of TGF-β1 in the lung tissue might be one of the mechanisms in treatment of radiation lung injury.
MDM2 gene and radiobiological effect in tumors

Abstract:
Murine double minute 2 (MDM2) gene is a eritieal eomponent of the responses to both ionizing and UV radiation. The level of MDM2 expression determines the extent to whieh radiation induees an inerease in the aetivity of the p53 tumor suppressor. MDM2 aets as a survival faetor in many eell types by limiting the apoptotie funetion of p53. In addition, expression of MDM2 is indueed in response to DNA damage, and the resulting high levels of MDM2 protein are thought to shorten the length of the eell eyele arrest established by p53 in the radiation response. Inereased levels of MDM2 appear to ensure that the aetivity of p53 returns to its low basal levels in surviving eells. Deereased levels of MDM2 sensitize eells to radiation. Thus, MDM2 is a potential target for therapeutie intervention beeause its inhibition may radiosensitize the subset of human tumors expressing wild-type p53 sueh that radiotherapy is more efficaeious.
Analysis of stereotatic radiotherapy for brain metastases

Abstract:
Objective To investigate the effect of stereotatic radiotherapy (SRT) for brain metastases. Methods Retrospect compare 66 patients treated by SRT with whole brain radiotherapy (WBRT+ SRT) to 26 patients treated by SRT alone, Results The patients treated by WBRT+ SRT had significantly benefit in lesions recurrence (13,3% vs 42.2%), middle recurrence time (10.5 months vs 4.2 months) and middle survival time(13 months vs 6.5 months) over the patients treated by SRT, respectively (P<0.05), But there are no different in terms of improving of Karnofsky score, effective rate, one-year survival rate, one-year death rate between the two groups. Conclusion WBRT+ SRT play an important role in treating brain metastases, by improving patients' life quantity and reducing recurrence.
Prognostics analysis of common radiotherapy combined stereotactic radiotherapy for non-small cell lung cancer of old patients

Abstract:
Objective To observe the effect of stereotactic radiotherapy (SRT) for non-small cell lung cancer(NSCLC) of old patients. Method 128 NSCLC old patients receive common radiotherapy and SRT. The results was followed. Results 1-year survival rate is 65.72% for all patients on univariate analysis radiation dose,tumor volume, Karnofsky performance status (KPS) are significantly associated with first follow-up response(FFR). On multivariate analysis only the first two are significant. Prognostic factors of survival time are FFR, tumor dose and clinical stage. Conclusions SRT can significantly raise survival time expecially for phase Ⅲ patients. The dose 72 Gy for NSCLC old patients is safe.

Abstract:
Objective Approach the radiology and clinical manifestation of pulmonary inflammatory pseudotumor. Methods Radiography and spiral volume scan for the lung of 41 patients. Results Many of the pulmonary inflammatory pseudotumor located in the surrounding of lung. According to the shape of CT, it included nodus (or lump) type and infiltrating type. The mark appearance included peach cusp sign, straight sign, net-spot sign, halo sign and uniform strengthen. It might he useful to the diagnosis when analyze the history and symptom before the shadow occurred in the lung. Conclusion CT imaging presented important value for the diagnosis of pulmonary inflammatory pseudotumor.
