重离子射线诱导肿瘤免疫原性的机制及其临床转化前沿

Mechanisms of heavy ion radiation-induced tumor immunogenicity and frontiers in clinical translation

  • 摘要: 重离子射线具有独特的物理剂量分布特征和出色的生物学效应,其不仅在局部肿瘤精准杀伤和控制方面表现出显著优势,还可通过DNA损伤应答等机制调控免疫相关分子及信号通路,激发肿瘤免疫原性并重塑肿瘤免疫微环境。笔者围绕基于重离子射线的新型放疗模式,系统阐释其免疫激活机制及相关分子特征,重点剖析重离子射线与免疫检查点抑制剂等免疫疗法的协同作用,总结最新的临床及临床前研究成果。基于重离子射线的新型放疗模式联合免疫治疗已经展现出巨大的临床转化潜力,将推动肿瘤治疗向更加个体化、精准化的方向发展,有望成为肿瘤放疗研究的前沿领域。

     

    Abstract: Heavy ion radiation is characterized by its unique physical dose deposition and superior biological effects. It not only offers significant advantages in precise local tumor killing and control but also modulates immune-related molecules and signaling pathways through mechanisms such as the DNA damage response, thereby enhancing tumor immunogenicity and reshaping the tumor immune microenvironment. The authors focus on novel radiotherapy approaches based on heavy ion radiation, systematically delineating their immune-stimulatory mechanisms and related molecular characteristics. Particular emphasis is placed on the synergistic effects between heavy ion radiation and immunotherapies such as immune checkpoint inhibitors, summarizing the latest clinical and preclinical research findings. The combination of the novel radiotherapy modality based on heavy ion radiation and immunotherapy has shown great potential for clinical translation. It will drive tumor treatment toward a more personalized and precise direction and is expected to become a frontier in tumor radiotherapy research.
